Abstract

Our study aims to optimize and establish measles bulk production procedures by usingplastic 10 layer cell factories measles
bulk production technology on 10 layer plastic cell factories based on transfered technology from Japan produced on glass Roux bottles. The results showed that SPF CEC can adhere and grow well on plastic bottle. Suitable density of cells was 118.000 cells/cm2., suitable MOI was 0,003. The volume of medium for cell culture: 3000ml, medium volume added
after virus inoculation and after changing were 2000ml. We already established measles bulk production procedure by using 10 layer plastic cell factories as follows: CEC culture in 2 days → washing cells, virus inoculation, adding medium and incubate at 32.5 ± 1oC in 3 days → changing medium and continue incubate 32.5 ± 1oC in 6 days→ keep cool 2 - 8oC overnight, harvest vaccine→ adding stabilizers, fltration → freezing quickly, distribution to 10 litter stainless steel tanks and stored at ≤-65oC. With improvement production procedures, we can increase yield upto 3.200.000 doses of fnished product per each bulk batch 30 plastic 10 layer cell factories, saving the cost, don’t have to use chemical to treat roux bottles and can recycle plastic cell factories for other purposes.
