Evaluation of PCR and LAMP techniques for detection of Burkholderia pseudomallei

In recent years, the record of cases of melioidosis caused by B. pseudomallei has increased signifcantly in the world. This is
explained by the increased disease spread and the improvements in accurately and rapidly detection of pathogen. This study aims to develop and evaluation two techniques PCR and LAMP (Loop-Mediated Isothermal Amplifcation) for detection of Burkholderia pseudomallei. Primers are designed based on the sequence ofr2 gene of type III secretion system (TTSS1). The isolated clinical B. pseudomallei strains have detected by PCR and LAMP techniques with a sensitivity of 10 CFU/ml. For DNA samples isolated from soil-mixed bacteria, the LAMP detection limit was 102 CFU/ml higher than PCR (104 CFU/ ml). Specifcity of PCR and LAMP are 100% and 95%, respectively. In the diagnosis of melioidosis, PCR can be useful in laboratory confrmatory methods, LAMP is suitable for rapid screening.
