Generation of norovirus antibody and development of an enzyme linked immunoassay for norovirus detection

Worldwide, human norovirus (NV) is an important cause of outbreaks and sporadic cases of acute gastroenteritis. Diagnostics for NV have been based on molecular method such as RT-PCR, real time RT-PCR and immunoassays. To date, several commercial enzyme immunoassay kits for detecting NV are available but the cost of these kits make them unsuitable for use as point-of-care test. In this study, we produced rabbit anti- norovirus antiserum using NV-GII.4 VLP as antigens. A sandwich ELISA to detect NV in fecal samples was established and validated. The results showed that the ELISA assay using NV-GII.4 antiserum has excellent specificity 100% and good sensitivity 82.0% (95% CI, 76.5 to 87.5), comparing to the gold-standard real-time RT-PCR for detecting NV-GII.4 strain, while other genotypes were not detected. The sensitivity of the assay was improved significantly for samples having Ct value lower than 21.4 (clinical cut-off value indicating NV is the cause of disease). Thus with the addition of antisera specific to other NV genotype, this assay could easily be applied for NV detection in clinical settings
